Hot line can mean inside information. Often hot lines are used for emergency assistance. Work my lady could mean any number of things. Without any context it is difficult to venture an educated opinion but I am guessing it means nothing related to kitchen work. Most likely it is something along the lines of romantic or sexual in nature.
Example: I need to know what my girlfriend wants from me emotionally. Your her brother, got any information?

I searched the lines in question and found them on an episode of 'Kitchen Confidential.'
Here is what they mean:
'I need to work my lady.': An old girlfriend of the head chef is in the restaurant. He wants to start over with her, so he is acting as her waiter. He is 'working her,' giving her special service, trying to charm her back into his life.
'You got the hot line?': In order to have the time to 'work his lady,' the chef must make sure others in the kitchen are taking care that things run smoothly. So he asks the other guy, 'You got the hot line?' which means 'Can you be responsible for the hot line?' The hot line is, I believe, the line of stations where food to be served hot is cooked.
