I have to make calls.

I have to make many calls.

Are these sentences appropriate?

They're both fine, but a native speaker would tend to say........

I have to make lots of calls.

and sometimes.......

I have to make loads of calls.

Wont it be better to say "I have to call many people" instead of "make calls"..?

You certainly should: it's a bit formal. People generally have lots of friends rather than many friends, there's lots of different newspapers and loads of football teams and so on.
