I cannot think of any occasion when you can use a/an in front of a superlative.

In your second example, most inspiring is not a superlative form. Most here means extremely.

I would add that we do not use the indefinite article with neither the superlative degree form nor adjectives nor anything else but singular countable nouns.
What is in front of the noun, it doesn't matter!?

That was a very, very, very big but ugly potato.

The phrase "an ugliest" would never work, but the phrase "a most inspiring" would, and does, but it doesn't contain a superlative construct: 'most' is an intensifier, meaning very.
