In my institute, there are 4 divisions. Grammar, Basic fluency, middle fluency and high fluency. If suppose I am in middle fluency,Can I say?
Before I was "in" Basic fluency. or
Before I was "at" basic fluency. here "in" or "at" which one is correct?

If you are talking about the class, which you are in your question, in.

If you are talking about the level of your command of the language, at.
