It means that we are ready to start or about to start something exciting/terrifying/adventurous/new. Depending on context, it might come across slightly differently but it basically means "We are about to start!" What we are about to start would only be clear from context.

emsr2d2 is right saying that more context would help. As far as I know, this expression is used, in addition to what emsr2d2 has said, when the speaker wants to express his/her reaction to something (positive or negative) that's just happened or is happening at the moment. For example, one can use this expression in a situation like this:
Mother: What do you think of marrying Bob?
Daughter: I don't like his hair.
Mother: Here we go. You know, I wonder if there is ever a guy that you'll ever approve of.
I was just thinking the same thing. Whilst I agree with 5jj's request to limit the number of punctuation marks, I realised that I would read a different context into it depending on the number of exclamation marks (rightly or wrongly used!)
(On a roller coaster) Here we go!!!!! = excitement.
(Talking to someone as in Bennevis' example) Here we go again! = annoyance
(At a restaurant) Here we go. = perhaps the arrival of expected food or something similar ("Aha. Here we go. Food's arrived.")
