Can I say "create a positive pressure in mines"? I feel very uncomfortable writing it. If it's not correct, how should I change it?

What do you want to say? You should provide more context.

At a push (...) you could 'create positive pressure [NB no article]' in a mine.People in the mining industry might say that - I don't know. I'd prefer something that specified what the difference was between: 'create a positive pressure difference between the inside and the outside of a mine. (That is, for me at least, what's positive is the difference or the balance, rather than the pressure itself.)

If it is understood that "positive pressure" is in relation to the outside of the mine, then I think it is fine to say "create positive pressure in a mine."
