There is no difference in meaning. Let's is a contraction of Let us, just like all the other common contractions in English.

We were taught in College that "Let us" is a formal way of saying "Let's" so the difference is only in what situation one would use either of them e.i. formal or informal, isn't that so?

Using the long forms rather than the contractions is more often used in formal situations, yes.

I'm not a teacher, but I write for a living. Please don't ask me about 2nd conditionals, but I'm a safe bet for what reads well in (American) English.

You can't use "Let us" to ask for permission. "Let us" is a suggestion about doing something.
"Let's go to the movies" means "Why don't we go to the movies?" or "How about we go to the movies?" or "I'd like to go to the movies. Do you think that's a good idea?" or "I propose/suggest that we go to the movies" (although we wouldn't actually say this last one often.)
You say "Let's" or "Let us" to the people who are "us"

However, I think I can see the point you're making. There is another way of using "Let us" which I didn't think of last time.
"Let us go to the movies" could mean "Allow/Permit us to go to the movies" if you were talking to your parents. So, it's a command when it's addressed to someone other than "us". In this case you can't use "Let's".
You can say to a policeman, "Let us out of this jail!" In this usage, you can't say "Let's out of jail."

    NOT A TEACHER

    (1) Some excellent teachers and non-teachers have given you wonderful advice.

    (2) May I share with you something my teachers taught me? They told me to forget about the formal vs. informal difference. They taught me these simple rules:

    (3) Use let's ONLY when you wish to make a suggestion:

It's so hot. Let's go to the beach, everybody!

We have a question about English grammar. Let's ask the great teachers at usingenglish.com for the answers.

That restaurant has the same menu every day. Let's not go there this week.
(Some people say "Let's don't ...." I respectfully suggest "not.")

    (4) Use let us ONLY for an order or demand:

We want to go to the beach, and you keep telling us that we can't. Why can't we?
Let us go to the beach! We demand that you let us go.

You tell us that we cannot leave the building until the police have checked everywhere for the bad guys who are hiding in the building. We understand. But why can't we call our families on our cellphones? Please let us call our families!

We children love you. Both of you have been fantastic parents! But, please, don't criticize the people we want to marry. Let us choose our own spouses.
