    Which sentence is correct ?

    a rural population of around one quarter the total population

    a rural population of around one quarter of the total population

    A rural population of around one quarter the total population
    A rural population of around one quarter of the total population


    The first is possible, but the second is more natural.

    With 'half', the 'of' is not essential; I recommend using it with all other fractions.
