Hello,
-It was not Mr. Brown, I saw yesterday at the shop.
-Your brother, I met yesterday, seems very sempathic.
I know we can't use 'that' with non-defining relative clauses. But, can we construct a sentence as above while using non defining relative clause? Must we always use 'who, whom' with non-defining?

This is a cleft construction, not a defining relative construction:
It was not Mr. Brown, I saw yesterday at the shop. 
It was not Mr. Brown I saw yesterday at the shop. 
It was not Mr. Brown that/who I saw yesterday at the shop. 
Non-defining relative construction:
Your brother, I met yesterday, seems very sympathetic. 
Your brother, who I met yesterday, seems very sympathetic. 
Correct spelling: sympathetic

No, you can't do that without a relative pronoun.
