If it really means "Don't touch me!", then why we don't just say " Don't you touch me again!" instead of saying "Don't you touch me no more!"

No, they are not totally different in meaning. Don't touch me no more means the same thing as Don't touch me again.

It's a variant of English using a double negative. (Some would say it's substandard English.) Some would consider you uneducated if you use language like that. Therefore, I wouldn't talk like that during a job interview, especially if you are trying to get a job as a college professor.
