When using 'I' or 'me' in a sentence as the subject.
Can one write; 'Sam drove Sara and I to the store.'?
Or should it be; 'Sam drove Sara and me to the store.'?

The second choice is the correct one.

The easiest way to test yourself is to make separate sentences out of it:

Sam drove Sarah to the store. Sam drove I to the store. Sam drove me to the store.

The sentence in italics should (hopefully!) sound wrong, telling you that "me" is the correct choice.
