You cannot use "question" with "want"
Anyway, you can say: What is the question? or What`s the question ? or What`s your question ...."

What does he want? You cannot use "wants" here because this is an interrogative sentence , but
He wants to go there [affirmative sentence]

It might be possible to use 'want' with 'question:
'Have you seen the essay question? Did you understand it?'
'I think that the question wants us to...'
(Mind you, this is stretching the point a bit- normally it wouldn't work)

Yes, you can add "s" to the verb "want" when it is in the 3rd person singular. present simple tense, affirmative:

e.g. He wants to go there.
She wants to tell you something.
"I think that the question wants us to..." [ as TDOL has already specified in his answer]
