Correct sentence.

Please explain which sentence is correct, and if possible explain it by giving some example.
i) The doctor ran a few test on me, and diagnosed me malayria.
ii) The doctor ran a few test,and diagnosed malayria in me.

secondly, some times, we come across text in which some words are written in italics for example "Indeed, a recurring 'leitmotif' throughout is the devastating impact ............."

I have written 'leitmotif' in inverted comma,actually,I read it in italics.And my italics key of my PC are not responding in this programme that's why I have written it in inverted comma, but this may be regarded as italics.

Neither sentence is correct. Here are some possibilities:

The doctor ran some tests on me and diagnosed malaria.
The doctor ran some tests and diagnosed me with malaria.
The tests that my doctor ran revealed that I have malaria.

Italics are used primarily for highlighting things. I have included a link explaining the use of italics.
