Honestly, I don't think I've ever heard of either "...I am giving you his regards..." or "He is giving you his regards...". The most common thing to say would be "You have regards from...[him, her, John]" About "Let him know" and "Give him this message", I guess I would say "John/Jane/Santa wants you to know that..." and "You have a message from John/Jane/Santa"... EDIT: Hmmm..Or would be.. I'm not so sure now even for "You have regards from". I think I would just say "Regards from"...Let's wait for more opinions, shall we..

Usually people say "he sends his regards". If you are passing on a message from another person, you could say "he asked me to tell you that...."

People also sometimes say 'remember me to Joe'; to deliver this message (very informally) you could say 'Jane asked to be remembered to you'. [I said 'very informally' because while the active remember me to ... is fairly common, the passive is relatively rare.] b

If you were being formal, you would say "Goober sends his regards." If you were being informal, you would say "Goober says hello." If you live in the American South, you would say "Goober says 'hey'". Unless you are talking to a head of state or corporate executive, it's more common to say "I just saw Goober the other day, and he aked me to say "hello".) Similarly, if you're passing along a message, you'd say something along the lines of: "Goober wanted me to tell you...." "Goober asked me to mention...."
