I have two brothers.

I'm an English teacher in France. I'm French. I have just received the last programms for teaching English. They say we can teach ' I have two brothers' but I'm not sure at all ! Can you tell me if this sentence is correct ? Or do I absolutely need 'got' : I've got two brothers. If it is correct, is 'I've two brothers' correct too ?

Both have and have got are used for possessions So, yes, these express the same meaning:

I have two brothers. / I've two brothers.
I have got two brothers. / I've got two brothers.

The verb have is used in many ways, including possession, whereas have got is only used for possession.

Yes, it's correct but less usual nowadays. It's better to say: I've got two brothers.;-)
