I would say that there is no real difference between the two.

"Knock at" suggests that you are knocking whilst stood at the door.

"knock on" suggests that you are knocking on the piece of wood that is the door.

The end result is the same in both cases.

I am British and I would use both "on" and "at" indiscriminately.

I have always heard "knocking on the door" used to mean physically hitting the door, but "knocking at the door" is hitting the door specifically to be let in, because you are waiting at the door. So if you were hitting the door while putting nails in it or just to make sounds, you would be knocking on the door but not at the door.
