2. The boy is playing with his toy with joy.
Which sentence is correct?

The first one isn't correct unless his toy is a flute or something. In any case, I would avoid using 'with joy'. So, 'The boy is joyfully playing with his toy' or 'The boy is happily playing with his toy.'
