2. He and two other partners work on the project.

He and the other two members work on the project.
I think the adjective,'other ' modifies 'two members' to mean two members other than him(He). Besides this, you can particularly point to some two members. I doubt whether we can imply the same meaning using the sentences mentioned by you.
I hope some senior member will tell us about this in detail.
