They are just one and the same expression. |   ... and the full, unabbreviated form allows you put a strong emphasis on the "does" - to add sarcasm and make a rhetorical question. So 'What's she look like?/'What does she look like?' is a simple question about someone's appearance; whereas 'What does she look like' isn't a question at all - it means 'I think she looks totally ridiculous'.
