Default Title of a letter

Hello, I wanted to know how to insert a title in the body of a formal letter.

If you are talking about the SUBJECT of the letter then it is usually placed after the salutation but before the body. LIke this:

Dear Sir,
Subject of the Letter here
Body of the letter here.....

You can use Re:

Dear Sir,
Re: Subject of the Letter here
Body of the letter here.....

Yes you may use "RE" but understand that "RE" stands for "reference" and is usually used in response to someone else's letter.

For example, if I write to you "Subject: Parts Order - Fred Smith" you might write back to me "RE: Parts Order - Fred Smith"

Do you mean how to format the title of a book or movie in the body of a letter?

If you're using a computer or word processing program, the standard format is to type movie, book, TV show or album titles in italics. Song titles are set in quotation marks.
