i think that you mean the relation between -er and the syllable of the stress.
first you should know that suffixes never chang the syllable of the stress they just the cause of the stress, but the prefixes are cause the syllable stress and the stress is between prefix and the base word. so if we have Base word + Suffix then we have stress on the first (e.g. reader) but if we have Prefix + base word + Suffix then we have stress on the second(e.g. unsafely).
