I can't say as I've ever heard a woman/girl use that, HKB but it sure is possible. Here are some more similar ones:

I gotta take a whiz/pee.

I gotta see a man about a horse.

Women would say more like:

"I have to tinkle"
"Time for a pee-pee break"
"I have to powder my nose"...
generally it is not quite the "lady-like" thing to say anything other than excuse me and just go to the bathroom!

Men are the ones that seem to make more of a production and advise people that they are going to go "shake a lizard", "see a man about a horse" (usually used when they are going outside to relieve themselves), and the "take a leak", "water the garden", etc. etc.
