The word "not" is only used as an adverb. It is most often used to negate verbs. Sometimes, "not" becomes part of a contracted verb: doesn't (does not), don't (do not), wouldn't (would not), etc.
I will not (won't) do that.
She is not (isn't) home.
The word "no" has several uses. As an adjective, it can modify a noun:
No evidence was found by the police.
He reached no conclusions.
As an adverb or interjection, "no" can be used to preface a remark, often in answer to a question:
No, I won't be coming to the party.
No! Stop saying that.
It can also be used with comparative adjectives:
He felt no better after the treatment.
