how can I differentiate between Do something and Make something easily?
You cannot. The usage is idiomatic:
Make a bed, do the laundry, make a mistake, do your hair, make amends, do the hula, make a breakthrough, do her dirty, make a mess, do your homework, make sure, do business, make time, do time, make hay, do lunch, make lunch. . .
