1. My bag is rather big.
2. My bag if fairly big.

And how to use rather and when? Because I have no idea how to use it.

They are close enough to being synonyms in this case - that is, when you use "rather" as an adjective.
He's rather tall = He's fairly tall.
That was rather rude = That was unnecessarily rude.
I think he's rather attractive. = I think he's quite attractive.
