frank by nature,
frank in nature,
or anything else?

"Frank by nature" seems more likely to me. In any case, Techstar, you didn't offer any sentences, so we can't comment on them.

ok so if I say about myself "I am frank by nature" will be correct,
and if I say about somebody,I have to say "He/She is frank in nature" is it correct, it has different meaning, or it become meaningless?

"I am frank by nature" is perfectly fine. However, "I am open and honest by nature" is better. (The word "frank" is not used much except in phrases like "Let me be frank with you".)
I don't know what "frank in nature" means since I am not used to that expression.

I did a Web search for "frank in nature", and I encountered mostly Indian websites. That explains things some, don't you think? (In any case, the two expressions mean the same thing.)
