To know someone is to be familiar with their face and/or their behavior. If you don't know someone, then you are not familiar with their face and their behavior.

In our example. Mrs. A tells her husband that she is no longer familiar with his behavior. His behavior is not what she expects.

So that means Mr.A is behaving different than Mrs.A has known about Mr.A?

That's right. There are two meaning:

I don't know you! (I don't recognize your face; you are a stranger)
I don't know you! (I don't recognize your behavior; your behavior is strange)

Could it mean that she doesn't want to see him anymore/she's through with him/she will ignore or avoid him from now on?
