In American English Blink is the reflex of closing both eyes for a fraction of a second.

Wink is the deliberate act of closing only one eye (as a signal).

In older English wink was used where we now use blink. That's why there are expressions like "Quick as a wink" or "In the wink of an eye" to mean very fast.
