Is this sentence grammatically correct? Isn't in the box as best as you can better?

No. If you make it "Spell the word in the box as best as you can", the reader will look for a word in the box.

You must spell the word as best you can. You must do that in the box. I don't see your problem.

Either sentence is correct, but one is better than the other because it is clearer.

I find 'as best as you can' very clumsy.
I would prefer:
'As well as you can' or 'The best you can'.

2. Come back as quickly as you can to the office.

Do both sentences sound good to you?

In most situations I'd probably say the first, but both are fine.
