In the movie, the interviewer asked:"what would you say if a man walked in here with no shirt, and I hired him?"
To which, Gardner answered "He must have had on some really nice pants."

What does that mean?

The reason he was hired was because of his fashion (his clothes), not his looks (his body).

He must have been wearing some really nice pants.
The joke is that, if the man was wearing really nice pants, that would make up for his not wearing a shirt!

I agree with Raymott.

I haven't seen the movie but it makes me think of the phrase "dress for success". This saying says that if you want to be successful, you have to first dress the part. In a sense, how you dress determines how successful you can be. Another idiom this makes me think of is "to lose your shirt" -- to lose your money. Shirt is used in some idioms about prosperity and generosity. So a guy with no shirt could be someone who's down on his luck or poor. But he still has to have nice pants...
