    1-There's this song that, when you sing it, soothes you incredibly.
    2-There's this song and when you sing it, it soothes you incredibly.
    3-There's this song that, when you sing it, you feel a lot better.
    4-There's this song and when you sing it, you feel a lot better.
    4-There's a song and when you sing it, it soothes you incredibly.
    5-There's a song that when you sing it, you feel a lot better.

    Which of the above are acceptable in informal spoken English?
    I don't think any of them would be acceptable in formal English.

    I don't think any of them would be acceptable in formal English.
    I'd say they were all acceptable in spoken English. You are more likely to say "feel" than "soothe" in informal usage though.
