Default When we use Are/Is and Do/Does in question form

I have a small question about questions.

When we use Are/Is and Do/Does?

    Examples:

Are you ready? ( to be )
Do you ready? ( present simple )

    Are you here?
    Do you here?

What is different between these parts of questions?
