I'd like to know the difference between "1" and "2".

    1. What's all that about?
    2. What's that all about?

In 2, you want to know everything about it as you don't know or haven't understood. To be honest, I don't think 'all' means much in the first, but it could ebe used to indicate that something is big; you could use it, say, if you heard people having a serious row.

Number one is something of a colloquial expression used in recent years (popularized by comedians like Jerry Seinfeld) to express confusion or incredulty at a series of events or circumstances:

We drive on the parkway and park on the driveway. The third hand on a wristwatch is called the "second hand." "Phonics" is not spelled the way it is pronounced.

    What's all that about??

It's another way of asking someone to explain the unexplainable.
