Is it "Your time and consideration is greatly appreciated" OR "Your time and consideration are greatly appreciated".

I'd use the plural, considering them two different things. You can also say "I greatly appreciate your time and attention."

    Re: Ending a cover letter

    Which is better:

I greatly appreciate your time and attention...

    or

I will greatly appreciate your time and attention...

In the sentence, you are talking about something which is yet to happen, right?

    Also, which is better:

I will great appreciate it if you would...

    or

I would greatly appreciate if you would...

Time is money. Assume that the person you are writing to is a highly paid individual whose time is valuable. Don't waste his time with something that is additional but doesn't add anything.

I think the 'it' is required.

Let's rearrange the clauses in the sentence.

If you........ , I would greatly appreciate. Does this sound right?

Context is key. Either of the first two sentences should probably be left out of a cover letter (especially the second one). The third one would never be used. (For one thing, great is an adjective, not an adverb.)

It's necessary. (Originally, I didn't notice its omission.) The word "it" stands for what would be appreciated.
