Many times in this forum and others, it has been noted that English does not have a "how manieth." You have to ask a longer question.

I'm the second oldest. Where do you fall in the birth order in your family?

Lincoln was the 16th president. How many presidents came before Lincoln? (Or maybe "What number president was Lincoln?")

By the way, no need to capitalize "second."

    What number son are you?

Not particularly elegant - there is no elegant way of posing this kind of question - but grammatical and meaningful.

I usually use a question : "How do you come in your family?" Are you the eldest?

For that, try "where" rather than "how" -- although the follow on question about "Are you the eldest?" makes it clear what you mean.

(American English for "where." That might not work elsewhere.)
