[STRIKE]Sir,[/STRIKE] This is unnecessary and offends the female teachers of this forum.

I want to know if this is a correct sentence.
It is not correct.

Perhaps you mean 'I will be taking up/resuming my duties on 16 April, 2016'.

Without more context, it's hard to say.

I would use the/your office, or say something like I will be starting work at your office on 16 April.

Without more context, it's hard to say.

But it's not too hard to say that join your duties is unlikely to be correct in any context- you can join meetings, etc, but not duties IMO.
