It is not correct.

Perhaps you mean 'I will be taking up/resuming my duties on 16 April, 2016'.

Without more context, it's hard to say.

I will be joining office on 16 April 2016

Dear Sir, Please correct the following sentence-

"I will be joining office on 16 April 2016."

I would use the/your office, or say something like I will be starting work at your office on 16 April.

But it's not too hard to say that join your duties is unlikely to be correct in any context- you can join meetings, etc, but not duties IMO.
