Hi,

1- It affects people like me who/that are used to this pain.

2- It affects people like me who/that is used to this pain.

Could you please tell me which one is the correct usage? In my opinion, both sentences are OK but I am not sure whether the 'that' in the first sentence is proper or not.

Use "who" when talking about people (general rule). After "people" you need to use "are" so sentence 2 is incorrect with "is".

It affects people like me - [people] who are used to this pain.
It affects people who are used to this pain - people like me.

Yes. Even though "who" is preferred by most, "that" can be used to refer to people.
