I always get myself confused with this rule.

One of the people who make me happy is Janet.

She is one of the people who make me happy.

Am I correct?

When shall I use the singular form, makes?

Yes, you are correct.
You don't use the singular with 'people'. You can say, "The person who makes me happy is Janet."

So in the sentence,"You're one of those who make me happy." Who is actually referring to those and not to one?
