    Most of students understood the lesson.

    and should I use "the" before "studens"

    I don't see a difference here:

    • Most of the students understood the lesson.


    You can omit 'of the'. Speakers know it's there; which is why they omit it now and then.

    That the plural noun students takes definite the has to do with the semantic scope of All. The determiner 'defines' the students within the scope of All. All, of, and the are semantically tied, which is why 'of the' is often omittable. That is, it's enough just to say All students because of the is recoverable in the meaning of All.
