I'm sure they both mean the same, but I would finish your sentence by saying, 'leave the rest for John'.

Thank you. One more question, can we use 'the others' to replace 'the rest' if there are quite a few cakes?

I think you can use either 'the rest' or 'the others'. They both mean 'what's left'.
