    The third sentence works logically if you have only one cousin. But the first is the natural expression.

    "She is my cousin" remains the natural way to express the fact.

    "She is a cousin of mine" states that she is a cousin and you have more than one cousin.

    "She is the cousin of mine" is not a natural utterance on its own.

    It is possible to begin a longer sentence like that, e.g. "she is the cousin of mine that went to college overseas" to distinguish this one cousin from among many. Even in that case "she is a cousin of mine..." is more likely.
