What does this sentence mean?

She was my keeper twice over—as don and as Mom’s eyes and ears.

When you say something happens twice over, three times over and so on, you are referring to the number of times it happens and emphasizing that it happens more than once.

In this context, I think it is to do with the fact that "she" is the writer's keeper "in two ways", not a specific number of times. She is the writer's keeper in two ways: 1) as don and 2) as Mom's eyes and ears.

Without context, I have no idea quite what that means but I think that's the gist of "twice over" here.
