Original sentence:Isabella requested the pope to draw a line of demarcation to separate Spanish finds from those of Portugal.

1.Isabella asked the pope for drawing a border line to separate Spanish findings from those of Portugal.
2.Isabella asked the pope for drawing a demarcation to separate Spanish findings from those of Portugal.

Does original sentence mean still the same as sentence 1 and 2 if I change the word "request" into the phrase "ask for" like sentence 1 and 2?

Yes, "request" means "ask for."

Your sentences need some help, though.

Isabella asked the Pope to draw a border line to separate Spanish discoveries from those of Portugal.
