What does the following question mean? If possible, please also give me a possible answer to the question..

I think the question should better pose like
Which moral issue best defines you?
The answer to the question is very much personalized. So it will vary from individual to individual.
You are correct. Both have the same meaning. But my understanding is that in this case "Which" more precisely defines one's requirement.

As another example, you can say, "What country are you from?" If there were a choice of three or four possible countries, you'd be more likely to say "Which country are you from?"
There are plenty of people who don't really care about any moral issues so they would be unable to answer that question (even if it made sense).
Remember - if you don't use correct capitalisation, punctuation and spacing, anything you write will be incorrect.
