It's due first thing Monday morning.

    M: When is it due?

W: It's due first thing Monday morning.

Does 'first thin' mean first class in this sentence?

No. It means early on Monday morning. In a work context, it means as soon as work starts in the morning.
I'm not a teacher of English, but I have spoken it for (almost) all of my life....
