    Some context, as usual.
Please be aware that I'm neither a native English speaker nor a teacher.

    Re: lack

'What do I lack?' is grammatically correct, but extremely unlikely. If you had supplied a context, you wouldn't have been misled by CS's unqualified 'Yes'. In many contexts, one would say 'What haven't I got?' or 'What don't I have?', or 'What am I missing'... (which, incidentally, tells you what lack means).

PS This context has occurred to me: if I'm going out, I might pause at the front door pat my pockets and say 'What have I forgotten?' If I said 'What do I lack?' MrsK might fear for my sanity.
