    ",...in spite of..." or "...despite of..." inside a text.

    What would be the correct option inside a text: ",...in spite of..." or "...despite of...". For example: "there is a large number of criticism is spite/despite of....."

    Despite means In spite of. There is no such thing as despite of.

    They are usually interchangeable, but your example 'sentence' is neither grammatical nor complete.
