    We usually say, "What is the story line in this movie/book."
    Can I used it in the TV show?

    or it should be "What is the show about?"

    You can certainly say "What is the story line in (I prefer 'of' to' in') this TV show" provided the show has a story line. Many TV shows have a theme but not a story line.
