    It was a relief to know that ...
    I feel a pain in my knee every time I try to run.

    could you explain me why I cannot say "With an encouragement from her older sister"?

    "Encouragement" is generally used uncountably.
    Remember - if you don't use correct capitalisation, punctuation and spacing, anything you write will be incorrect.

    I prefer "With hope in my heart, I went off to meet the object of my desire" (no article before "hope").

    "It was a relief to know that ..." is only grammatically correct with the article.

    "I feel a pain in my knee ..." and "I feel pain in my knee ..." are both possible. More natural would be "My knee hurts every time ...".

    Many words can be used both countably and uncountably. You can use pain uncountably for something general and a pain for a specific pain.
