Which of the following is correct and if both are correct ,do they have same meaning?

1:- He is the best player in the team.
2:- He is the best player of the team.

Neither one is correct - the correct preposition here should be 'on'.

He is the best player on the team.

Yes. '...in the team' is fine in BE.

Hmm, always interesting to hear and compare differences in the different English variants. I'll gladly amend my original comment to 'in the team' is completely unacceptable in AmE only, then.

"In the team" sounds very odd to me also.

They're all fine and mean the same thing. None will make anyone faint. But I agree with Skrej that in the U.S. on is the most natural.
